2-Chloro-4-amino-5-methylpyridine CAS number is 79055-62-2, molecular formula: C6H7ClN2, molecular weight: 142.588, The existing synthesis technology of 2-Chloro-4-amino-5-methylpyridine generally uses 2,3,5-trimethylpyridine as raw material, which is oxidized, nitrated, methoxylated, rearranged in acetic anhydride, alkaline hydrolyzed, chlorinated. The sulfoxide is chlorinated to give the final product 2-Chloro-4-amino-5-methylpyridine CAS number 79055-62-2. This method can be used for industrial production, but it is not ideal compared to another method.
The preparation process of the more optimized technical scheme of 2-chloro-4-amino-5-methylpyridine is as below:
1. In a 2 L flask, 145 g of 2-chloro-5-methylpyridine, 1200 mL of purified water, and 12.8 g of sodium tungstate were added in one portion. 18.4 g of concentrated sulfuric acid was added dropwise, and the mixture was stirred for 30 minutes after the addition. Then, the temperature was raised to 65-75 ℃, 320 g of 30% H 2 O 2 was added dropwise, the temperature was controlled at 65-75 ℃, and the dropping time was controlled at 5-6 h. After the completion of the dropwise addition, the reaction was kept at a constant temperature for 40 hours, and TLC was monitored. When the reaction was not progressed, the temperature was raised to 95-100℃, and the reaction was continued for 10 hours. After the reaction was completed, it was cooled to room temperature, the pH was adjusted to 8-9, and the unreacted raw material was extracted with 500 mL of toluene, followed by extraction of the product 4-5 times with dichloromethane. Dry and spin dry and air dry to give a dark yellow solid product 145g.
2. Add 50 g of the dark yellow solid obtained in the step 1 to the flask, add 44 mL of concentrated sulfuric acid at a low temperature, then raise the temperature to 70℃, and add the mixed acid formed by 58 mL of concentrated sulfuric acid and 48 mL of nitric acid. After the dropwise addition, the reaction is kept for about 3 hours. The reaction was monitored by TLC. After the reaction was completed, it was poured into ice water, stirred, filtered, and washed with ice water three times, and dried to yield 39 g of pale yellow solid.
3. In the flask, 80% ethanol (107 mL) and 30 g of the pale yellow solid obtained in Step 2 were heated to 60℃, HCl was added, then Fe powder was added in portions, the temperature was controlled at 60-65 ℃, and after reflux, the reaction was refluxed for 30 min. Left and right, the reaction was followed by TLC. After completion of the reaction, it was cooled to room temperature, adjusted to pH = 8 with NaOH solution, then filtered and dried. After heating to reflux with 4 volumes of dichloromethane, the mixture was cooled to room temperature, and petroleum ether was added to give a white product (17 g). This white product is 2-chloro-4-amino-5-methylpyridine CAS 79055-62-2.
