Product Description

3-amino-4-methylpyridine CAS number is 3430-27-1, his synthesis method can be roughly divided into the following according to its starting materials:
(1) using 2-amino-4-methylpyridine as a raw material, first diazotizing and hydrolyzing to obtain 2-hydroxy-4-methylpyridine, and then nitrating, phosphorus oxychloride and pentachlorobenzene with a mixture of nitric acid and sulfuric acid. Oxygen phosphorus chlorination, Pd/C catalytic hydrogenation reduction dechlorination to give 3-amino-4-methylpyridine cas 3430-27-1. The method has long route, low product yield, large amount of waste acid and waste water, serious environmental pollution, and is not suitable for industrial production.
(2) condensed with ethyl acetoacetate and cyanoacetamide to obtain 2,6-dihydroxy-3-cyano-4-methylpyridine, which is then chlorinated, hydrolyzed and Hoffman degraded to obtain 2,6-di Chloro-3-amino-4-methylpyridine is dehydrochlorinated by Pd/C hydrogen to give 3-amino-4-methylpyridine. Although this method avoids the disadvantages of traditional nitrification, it still has the disadvantages of low product yield and long reaction route.
(3) using trifluoroacetic anhydride as solvent, nitrating with concentrated nitric acid, and moving the nitro group to obtain 3-nitro-4methylpyridine by aqueous solution of sodium metabisulfite, and then reducing it by Pd/C catalyst to obtain 3-amino-4methyl group. In the first two steps of the method, the reaction time is long, and trifluoroacetic anhydride which is expensive and cannot be recycled is used in the reaction; therefore, industrial production is difficult to achieve.
(4) 3-bromo-4-methylpyridine and boronic acid ester are subjected to Grignard reaction to obtain 4-methylpyridine-3-boronic acid, then inorganic amide is used as ammonia source, and metal oxide is used as catalyst in solvent. A one-step reaction produces 3-amino-4-methylpyridine cas number 3430-27-1. The method has mild reaction conditions and high product yield, and is favorable for industrial production.
