The other one is methanol and 4-aminopyridine-3-carboxylic acid as a raw material, and a 4-aminonicotonic acid methyl ester is obtained by a single reaction, and the yield is about 68%. The cost of production through the two methods of cost accounting is roughly the same.

For example, 30 g of methyl 4-aminonicotinate was dissolved in a mixture of glacial acetic acid and water, and 52 g of liquid bromine was added dropwise at 0℃, gradually added to room temperature and stirred overnight. The reaction mixture was filtered, and the residue was washed with water and methyl t-butyl ether, and dried to give 27.7 g of 4-amino-5-bromo nicotinic acid. The obtained 4-amino-5-bromonicotinic acid was placed in a round bottom flask, and 200 mL of POCl3 was carefully added thereto, gradually warmed to 90℃, and stirred for 2 hours. The mixture was cooled to room temperature, then evaporated. The obtained 29.7 g of a solid was dissolved in 100 mL of anhydrous tetrahydrofuran, and then added dropwise to a solution of 19.3 g of NH4SCN in 200 mL of anhydrous tetrahydrofuran, and stirred at room temperature overnight. Filtration, filtration of the residue with water and drying gave 8-bromo-2-mercapto-2,3-dihydropyridine [4,3-d]pyrimidine-4(1H)-one. it is a key intermediate in the synthesis of many anti-tumor drugs.
